J-147 Powder is a phenylhydrazine compound with broad-spectrum neuroprotective activity with EC50 values of 60-115 nM. The compound decreased Aβ40 and Aβ42 levels and increased BDNF levels in the hippocampus. In wild-type rats, J-147v can enhance memory; And in genetically modified Alzheimer's mice, it prevented memory impairment and salvaged cognitive deficits.

In this study published in the journal Aging Cell, scientists confirmed that J147 binds to a protein in mitochondria (the energy factory of cells), making aging cells, mice, and fruit flies appear younger.

In 2011, after screening compounds from plants with the ability to reverse aging cells and molecular signals in the brain, research showed that J-147 powder can reverse memory deficits, enhance the production of new brain cells, and slow or reverse the progression of Alzheimer's disease in mice.

In this new study, its molecular target, a mitochondrial protein called ATP synthase, was ultimately discovered.

Previously, the enzyme had been proven to control the aging of nematodes and fruit flies, and this new study suggests that by manipulating the activity of ATP synthase, scientists can protect neuronal cells from the multiple toxicities associated with aging the brain.

Further experiments have shown that regulating the activity of ATP synthase with J147 alters the levels of various other molecules, including ATP itself, thereby making mitochondria healthier and more stable during aging and disease processes.

These results not only support further exploration of J147 as a treatment for Alzheimer's disease but also indicate that J147 may be useful for other age-related diseases.
