Product Introduction

Low-substituted hydroxypropyl cellulose (L-HPC) is a versatile non-ionic cellulose derivative primarily used as a disintegrant and binder in solid dosage forms. Due to its high surface area and porosity, its powder can rapidly absorb water and swell. When used in tablets, it facilitates rapid disintegration while its rough structure provides significant interlocking with drugs and particles, enhancing tablet hardness without affecting disintegration. As a result, it accelerates drug dissolution and improves bioavailability.

This product is a low-substituted hydroxypropyl cellulose ether, a versatile non-ionic cellulose derivative primarily used as a disintegrant and binder in solid dosage forms. Due to its high surface area and porosity, the powder rapidly absorbs water and swells, facilitating the quick disintegration of tablets. Its coarse structure allows for significant interlocking with drugs and particles, resulting in improved tablet hardness without affecting disintegration, thereby accelerating drug release and enhancing bioavailability. The recommended dosage of this product is typically around 2-5%. It can be incorporated internally during wet granulation in tablet manufacturing or used as an external addition. For optimal effectiveness, it can be added both internally and externally simultaneously.

Low Substituted-Hydroxypropyl Cellulose(L-HPC)

Low Substituted-Hydroxypropyl CelluloseL-HPC

1. L-HPC is mainly utilized as a disintegrant and binder in wet or dry granulation for tablet and granule formulations.

2.LH-11 is employed as a disintegrant to prevent tablet capping and as a disintegrant for direct compression tablets. LH-21 is used as a binder and disintegrant in wet granulation for tablet formulations.

3. Generally, the concentration of L-HPC in prescriptions ranges from 5% to 50%.

L-HPC and HPC:

HPC and HPC have different properties. HPC is extensively used as a binder in solid dosage formulations. L-HPC and HPC are discussed in separate chapters in pharmacopeias. Conventional HPC contains a significant amount of hydroxypropoxy groups on the cellulose backbone, while L-HPC has only a small amount of hydroxypropoxy groups . This chemical distinction results in HPC being soluble in water, whereas L-HPC is insoluble. HPC aqueous solutions are commonly employed as binders in wet granulation, but L-HPC cannot be used in the same manner. L-HPC, due to its water-swelling properties, is an effective disintegrant. Conversely, HPC does not exhibit this property. Moreover, L-HPC possesses good compressibility, allowing for the preparation of tablets with comparable hardness to those made with microcrystalline cellulose through dry blending. In this application, L-HPC acts as a "dry binder."

Focused on improving the compaction issues of solid dosage forms

1. Primarily used as a disintegrant and binder in solid dosage formulations

2. The powder possesses a large surface area and porosity, significantly enhancing moisture absorption speed and swelling, thereby accelerating tablet disintegration, facilitating drug dissolution, and improving bioavailability

3. The rough particle structure exerts strong interlocking effects, enhancing tablet hardness and glossiness, and improving the compaction efficiency and effectiveness of poorly compactable drugs

4. The recommended usage concentration in formulations is 5%-50%

Special applications of the product

1. Capping is a common problem during tablet compaction. Many reports indicate that capping is caused by excessive wall residues during the compression process. L-HPC can reduce the resistance and demolding force of wall residues during tablet compression.

2.LH-11 is a multifiber grade and has the best anti-capping effect. Its fibers intertwine effectively to prevent capping.

3. Compared to the three major super disintegrants (sodium starch glycolate, cross-linked sodium carboxymethyl cellulose, cross-linked polyvinylpyrrolidone), L-HPC has certain advantages in terms of water absorption rate and water absorption capacity. It does not gelatinize and does not cause "mottling" after the removal of compression stress.

4. Due to its preparation from high-quality wood pulp etherification, low-substituted hydroxypropyl cellulose exhibits strong chemical inertness and high compatibility with active pharmaceutical ingredients in formulations. It is commonly used in the formulation screening for cardiovascular and digestive system drugs.
